NMN Supplementation Protocol

Boosting NAD+ levels through NMN (nicotinamide mononucleotide) supplementation.

By David Sinclair

Overview

David Sinclair's research has highlighted the importance of NAD+ in cellular health and aging. NMN is a precursor to NAD+ and supplementation may help maintain NAD+ levels that naturally decline with age. Sinclair himself takes NMN daily as part of his longevity regimen.

How to Follow This Protocol

  1. Step 1: Choose Quality NMN

Select NMN from a reputable manufacturer with third-party testing. Look for >98% purity and proper storage conditions (some NMN is more stable refrigerated).

Note: Sublingual or liposomal forms may have better absorption
  2. Step 2: Start with Lower Dose

Begin with 250mg per day for the first week to assess tolerance. Some people experience mild flushing or GI discomfort initially.

    1 week
  3. Step 3: Increase to Target Dose

Gradually increase to 500mg-1g daily. Sinclair reportedly takes 1g per day. Take in the morning to align with circadian rhythms.

Note: Can be taken with or without food
  4. Step 4: Monitor and Adjust

Pay attention to energy levels, sleep quality, and overall wellbeing. Consider periodic blood tests for NAD+ levels if available.

Benefits

  • May increase NAD+ levels
  • Potential improved energy metabolism
  • Supports sirtuin activation
  • May improve mitochondrial function

Risks & Considerations

  • Limited long-term human studies
  • Quality varies between manufacturers
  • Potential interactions with other supplements

Contraindications

Do not follow this protocol if any of the following apply:

  • Pregnancy or breastfeeding
  • Active cancer (consult oncologist)
  • Those on blood thinners
